The Asian elephant (Elephas maximus) is an endangered species facing a decline in population, primarily due to Elephant Endotheliotropic Herpesvirus haemorrhagic disease (EEHV-HD), which is the leading cause of calf mortality. This acute haemorrhagic syndrome results from extensive endothelial destruction and disseminated intravascular coagulation, often leading to sudden death. This research investigates the impact of EEHV-HD on the European captive Asian elephant population, examining hereditary or zoo-associated risk factors for disease development, the coagulation status of healthy Thai and European populations, and potential genomic mutations in coagulation factor VII that may cause hereditary coagulopathy. Findings indicate that over half of captive-born fatalities were due to EEHV-HD, suggesting a link to zoo-associated factors and possible sire and dam influences. A specific point-of-care analyser established reference coagulation parameters, including PT, aPTT, and fibrinogen levels, categorized by gender, age, region, and EEHV status. These methods can assist in routine health checks and emergencies during EEHV-HD outbreaks. Additionally, new single point mutations in the coagulation F7 gene were identified in elephants from Thailand and Europe. Overall, the study underscores the need for ongoing epidemiological research to identify risks and protective factors influencing calf susceptibility to EEHV-HD.
